Glioblastoma (GBM), the deadliest form of brain cancer, long gift for localization issues. Chimeric antigen receptor (CAR) T cell immunotherapy has emerged as a potent strategy for treating cancer. IL-13-receptor-α2 (IL13Rα2), present in more than 75% of GBMs, has been recognized as an attractive candidate for anti-glioblastoma therapy. Here, we propose a multidisciplinary approach new for brain tumor targets using a combination of fluorescent, nanoparticle therapy and cell CAR T modified by the target-quadruple-mutant of IL13 (TQM-13) proved to have a binding of high affinity for IL13Rα2-expressing glioblastoma cell toxicity off low-target.

Knee osteoarthritis (KOA) is a common disease, especially in older people. This study investigated the expression of matrix metalloproteinase-13 (MMP-13), NF-κBp65 and interleukin (IL) -lβ in synovial tissue of patients KOA and the role of MMP-13 and NF-κBp65 signaling pathway in the pathogenesis KOA.
A total of 100 patients were enrolled KOA in our hospital from December 2015 to December 2017 and classified as either mild group KOA (Outerbridge grade 1 and 2) or a group of severe KOA (Outerbridge grade 3 and 4). non-OA patients included as controls. synovial tissue of patients in both groups were collected to detect mRNA and protein expression of MMP-13, NF-κBp65 and IL-lβ. Synovial tissue sections were subjected to hematoxylin and eosin and immunohistochemistry (SP method). cartilage tissue was observed under light microscope after staining safranin O-fast green.
Analysis of quantitative reverse transcription-PCR and Western blot showed that the expression of MMP-13, NF-κBp65 and IL-lβ in mild and severe group was substantially deregulated compared with the control group (all P <0.05). A positive correlation between MMP-13 and NF-κBp65 expression in synovial tissue identified KOA (P <0.05). Immunohistochemistry revealed that the expression of MMP-13 and NF-κBp65 associated with the severity of KOA (MMP-13: heavy, 92.54%; moderate, 76.52%; control: 32.14%; and NF-κBp65: heavy, 85, 56%; moderate, 48.12%; control: 28.32%).
This evidence suggests that the severity of KOA associated with the expression of MMP-13 and NF-κBp65.
